Introduction:
rhEGF is a soluble non carbohydrate protein, which contains 53 amino
acids. The composition, sequence and peptide map of rhEGF are strictly
accord with the natural hEGF. The Amino acid sequence of the first
five N-terminal amino acids was determined and was found to be Asn-Ser-Asp-Ser-Glu,
which agrees with the sequence of native human EGF. N-terminal methionine
has been completely removed enzymatically.

Expression
and purification system:
With the application of a high-effective secretive expression system
of Escherichia coli, target protein was secreted directly into the
culture medium after membrane modification. Extremely high bioactivity
achieved due to the remaining of advanced protein structures. By using
full sets of premium separation and purification system, purity of
the production reached above 95%, and the highest can be above 98%.
Application:
rhEGF can promote the treatments of the wounds of skin and cornea,
and has the potential of repairing wounds in alimentary tract, kidney,
liver, lung and trachea. It can be used in the treatments of burns,
all kings of surgical and postoperative wounds and the injury or ulcer
of cornea. Recommended concentration - 0.1-1.0 ng/ml medium depending
on cell type
rhEGF
can enhance the growth of new epidermal and stem cells, promote the
cell's metabolism and converse differentiation. It is widely used
in cosmetics such as whitening, anti-crinkle, crinkle dispelling,
anti-aging, etc.
Biological
Activity:
ProSpec's recombinant Human Epidermal Growth Factor is fully biologically
active when compared to standards. The ED50, calculated by the dose-dependant
proliferation of murine BALB/c 3T3 cells (measured by 3H-thymidine
uptake) is less then 0.1 ng/ml corresponding to a specific activity
of 1 x 1000000 Units/mg.
Storage:
2-8¡æ(in refrigerator) storage, stable within 36 months.
Endotoxin:
Less than 0.1 ng/¦Ìg (IEU/¦Ìg) of recombinant EGF.
Protein
content:
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy
at 280 nm using the absorbency value of 2.858 as the extinction coefficient
for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE
computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of human EGF as
a Reference Standard.
